Human Nectin-1 is a type 1 integral membrane glycoprotein that can form a homo-cis dimer on the cell membrane. The recombinant Nectin-1 dimer protein (CSP-25195-03) is a cis-homodimer (cis-dimer) and contains a Nectin-1 extracellular domain (UniProt# Q15223, amino acids Gln31-Ala355) fused with a proprietary dimer motif followed by a tandem His-Avi tag at the C-terminus. This dimeric protein is expressed in HEK293T cells. The recombinant human Nectin-1 protein dimer is bioactive and can bind to Nectin-3 and Nectin-4. It also binds Nectin-1-specific antibodies. This Nectin-1 dimer can be used as an antigen for in vitro assays and antibody screening, and as an immunogen for immunization to generate antibodies targeting more conformational epitopes.

Background
Human Nectin-1 is a type 1 integral membrane glycoprotein that can form a homo-cis dimer on the cell membrane. Nectin-1 is also known as Poliovirus receptor-related 1 (PVRL1), cluster of differentiation 111 (CD111), and herpesvirus entry mediator C (HVEC). Nectin-1 contains an extracellular domain with three immunoglobulin-like (Ig-like) domains. Nectin-1 interacts with both Nectin-4 and Nectin-3. Nectin-1 and Nectin-3 are expressed in neuronal tissue and unlike many other cellular adhesion molecules, they do not distribute evenly. Nectin-1 is primarily found on the axonal side and Nectin-3 is primarily found on the dendritic side. The interaction of Nectin-1 with Nectin-3 has been found to be essential for proper morphogenesis of the eye. Nectin-1 also functions as the entry receptor for Herpes Simplex Viruses 1 and 2 through interaction with glycoprotein D on the viral envelope. Therefore, a recombinant protein mimicking the Nectin-1 dimer conformation can be crucial for therapeutic discovery.
